Significantly expanded T cell receptor (TCR) Vβ-expressing CD4 T cells at baseline (pre-Tx) and during follow-up after haematopoietic stem-cell transplantation (after Tx), analysed with 22 TCR Vβ-specific mononuclear antibodies (IOTest Beta Mark, Beckman Coulter Immunotech) and flow cytometry
TCR | pre-Tx | 3 months after Tx | 6 months after Tx | 8 months after Tx | 12 months after Tx | 24 months after Tx | 36 months after Tx |
---|---|---|---|---|---|---|---|
Vβ 4 | 2.6 | 1.9 | 2.2 | 2.6 | 1.8 | 2.3 | 1.8 |
Vβ 12 | 2.1 | 3.4 | 3.5 | 3.2 | 2.5 | 1.7 | 2.2 |
Vβ 13.1 | 5.2 | 12.9 | 5.1 | 4.7 | 4.8 | 5.1 | 5.9 |
Vβ 13.6 | 2.4 | 1.3 | 2.4 | 1.9 | 2.4 | 0.7 | 2.8 |
Normal ranges were established for each Vβ member based on confidence intervals of 97.5% determined in 20 healthy controls. Perturbations of TCR Vβ families were considered to be significant (bold figures) when they were outside these normal intervals.
