However, the catalytic domain of Pol ε is dispensable for viability (Kesti et al., 1999), and Pol δ synthesizes the leading strand during SV40 replication (Prelich and Stillman, 1988). Consistent with this, an alternative model recently has been proposed suggesting that Pol δ synthesizes both strands at the replication fork (Johnson et al., 2015).

It is expressed throughout the entire cell cycle but the protein levels are thought to be highest during S-phase and lowest in the G2/M-phases (Celis and Celis, 1985; Takahashi and Caviness, 1993; Kawabe et al., 2002).
