Selective Deimination of Vimentin in Calcium Ionophore-Induced Apoptosis of Mouse Peritoneal Macrophages

Abstract

We found citrulline-containing proteins in mouse peritoneal macrophages undergoing calcium ionophore-induced apoptosis. Such proteins were products of deimination of arginine residues catalyzed by endogenous peptidylarginine deiminase (EC 3.5.3.15) activated by calcium influx. Western blotting analyses of the extract from macrophages incubated with 1 μM ionomycin showed selective deimination of vimentin without detectable degradation. Double immunofluorescence staining of deiminated proteins and vimentin suggested localization of deiminated vimentin around the periphery of round-shaped nucleus, which was thought to be an early morphological sign of apoptosis. The biological implication of vimentin deimination in macrophage apoptosis is discussed.

    A paradox in the study of PADs is that these enzymes require micromolar to millimolar amounts of calcium to achieve full activation in cells and in vitro [18,31]. Intracellular activation of PADs under these high calcium concentrations is, however, only observed during unique conditions, which also lead to cell death, such as keratinocyte differentiation, cytolysis induced by host or microbial pore-forming proteins, and artificially induced by calcium ionophores [13,31–36]. PADIs have key roles in nerve growth, onset of inflammation states, embryonic development, tissue aging, epithelial terminal differentiation, trauma apoptosis and transcriptional regulation of gene expression and regulation [3–10]. Furthermore, several human diseases such as Alzheimer's disease, rheumatoid arthritis, multiple sclerosis, psoriasis and different types of cancers are associated with the presence of PADIs and their citrullinated targets [9,11–13].
