Background To investgate the expression of Th9 in the peripheral blood and the level of IL-9 in serm of rheumatoid arthritis (RA)
Objectives analysis the relationship between this cells and disease activity, bone erosion, etc. and underlying the possible immunological mechananism of RA.
Methods We choose 36 case of patients and 22 case of normol control. Patients were divied into two groups: heavy,medium.We obtained PBMCs from subjects and patients, and those cells were defined as CD3+CD8-IL-9+(Th9) by CD8FITC, CD3APC, IL9PE in peripheral blood mononuclear cells from RA patients and healthy controls by using Flow cytometery. Detect the level of patients and healthy controls serum IL9 by ELISA test.And then to analyzed the correlation between the cells Th9,IL9 and clinical data of RA, such as ESR,CRP,RF, the number of tender joints,the number of swollen joints, morning stiffness and bone destruction, etc. While to analyzed the correlation between Th9 and level of IL9.
Results The expression of CD3+CD8-IL-9+cells (Th9) in PBMCs of RA was significantly higher than normal controls (0.989±0.498 vs 0.213±0.084, P<0.01).Meanwhile, the expression of Th9 in heavy activity of group were higher than those medium (1.181±0.523 vs 0.686±0.254,P<0.01). The level of Th9 in the two groups were all significantly higher than controls (P<0.01). There was a positive correlation with Th9 and DAS28 (r=0.461, P=0.005), ESR (r=0.347, P=0.042), CRP (r=0.384, P=021), TJC (r=0.341, P=0.042), SJC (r=0.347, P=0.038) and RF (r=0.379, P=0.025), the level of IL-9 (r=0.461, P=0.005).There were no relationship with Th9 and course,morning stiffness,CCP,the abnormal of EKG,bone erosion and vasculitis.In the four groups.The level of IL-9 was higher than normol control (1.055±0.421 VS 0.686±0.134,p<0.01).The serum level of IL-9 were decrease in turn (1.367±0.470, 0.939±0.301, 0.784±0.133, 0.686±0.134,each two groups was extremely significant (P<0.05). There was a positive correlation with IL-9 and DAS28 (r=0.385, P=0.048), ESR (r=0.403, P=0.018), TJC (r=0.420, P=0.023), SJC (r=0.361, P=0.035). There were no relationship with Th9 and course, morning stiffness, CRP, CCP, RF, the abnormal of EKG, bone erosion and vasculitis (P>0.05).
Conclusions High expression of Th9 and level of IL9 maybe through the whole disease process, induce disease activity. These results indicate that the abnormality of Th9 may play an important role in the pathogenesis of RA.
